We still don't know whether two transcription pre-initiation complexes (PICs) can co-assemble at disparate locations or what structural interactions might result from that. We explored the assembly of the Saccharomyces cerevisiae Mediator-PIC on a divergent promoter to look into the structural roots of this event.
We were able to observe two separate PICs co-assembled at divergent promoters using data from Cryo-EM, crosslinking-mass spectrometry, and in vitro transcription experiments, which is compatible with a recently developed concept drawn from in vivo studies.
Overall, the structure of the Mediator-PIC in this unique form is favorable for differential transcriptional activation.
