For leading and lagging strand DNA replication in yeast, and most likely in other eukaryotes, there exist separate DNA polymerases. While DNA polymerase delta () is in charge of producing Okazaki fragments and lagging strand replication, DNA polymerase epsilon () is predominantly in charge of leading strand replication.
Multiple unique enzymatic activities are required for the creation of each Okazaki fragment (1). The error-prone DNA polymerase (Pol) extends the short RNA primer that is created by the primase component of the Pol-primase complex (2,3).
