The correct answer would be the second one, "his antagonistic relationship with his father". Kafka's father wanted him to become a businessman, when all he wanted to do and be was a writer. Than infuriated his father, which led to their antagonistic relationship. It is reflected in the story as well - Gregor's father is very short-tempered, disappointed with his son, just as Kafka's father was.
